SAN FRANCISCO — Moderna Therapeutics, the most highly valued private company in biotech, has run into troubling safety problems with its most ambitious therapy, STAT has learned — and is now banking on a mysterious new technology to keep afloat its brash promise of reinventing modern medicine.

Exactly one year ago, Moderna CEO Stéphane Bancel talked up his company’s “unbelievable” future before a standing-room-only crowd at the annual J.P. Morgan Healthcare Conference here. He promised that Moderna’s treatment for a rare and debilitating disease known as Crigler-Najjar syndrome, developed alongside biotech giant Alexion Pharmaceuticals, would enter human trials in 2016.

It was to be the first therapy using audacious new technology that Bancel promised would yield dozens of drugs in the coming decade.

But the Crigler-Najjar treatment has been indefinitely delayed, an Alexion spokeswoman told STAT. It never proved safe enough to test in humans, according to several former Moderna employees and collaborators who worked closely on the project. Unable to press forward with that technology, Moderna has had to focus instead on developing a handful of vaccines, turning to a less lucrative field that might not justify the company’s nearly $5 billion valuation.

“It’s all vaccines right now, and vaccines are a loss-leader,” said one former Moderna manager. “Moderna right now is a multibillion-dollar vaccines company, and I don’t see how that holds up.”

Bancel made no mention of the Crigler-Najjar drug when he spoke Monday before a similarly packed room at this year’s J.P. Morgan conference.

His presentation instead focused on four vaccines that the company is moving through the first phase of clinical trials: two target strains of influenza, a third is for Zika virus, and the fourth remains a secret. Bancel clicked through graphs of data from animal studies before hurrying on to tout Moderna’s balance sheet and discuss the company’s cancer vaccines, slated for clinical testing later this year.

When STAT asked Bancel after the presentation about Crigler-Najjar, he deferred to Alexion.

Founded in 2012, Moderna reached unicorn status — a $1 billion valuation — in just two years, faster than Uber, Dropbox, and Lyft, according to CB Insights. The company’s premise: Using custom-built strands of messenger RNA, known as mRNA, it aims to turn the body’s cells into ad hoc drug factories, compelling them to produce the proteins needed to treat a wide variety of diseases.

But mRNA is a tricky technology. Several major pharmaceutical companies have tried and abandoned the idea, struggling to get mRNA into cells without triggering nasty side effects.

Bancel has repeatedly promised that Moderna’s new therapies will change the world, but the company has refused to publish any data on its mRNA vehicles, sparking skepticism from some scientists and a chiding from the editors of Nature...

mRNA-1647 is the "bioequivalent" GTMP all of the Moderna regulatory paperwork was done with, and it's been around since 2017. Moderna had mRNA tech ready to go five years ago.

absolutely incredible. they had this stuff written up since 2017, just waiting on the right pandemic. mrna-1647 could be the placeholder for mrna-1273, covid 2p-S, or maybe mrna-1192, ebola glycoprotein, etc? these screenshots are both p302

Judicial Watch: FDA Records Show Significant Number of mRNA Test Rats Born with Skeletal Deformations

(Washington, DC) – Judicial Watch announced today that it received 699 pages of records from the Department of Health and Human Services (HHS) regarding data Moderna submitted to the Food and Drug Administration (FDA) on its mRNA COVID-19 vaccine, which indicate a “statistically significant” number of rats were born with skeletal deformations after their mothers were injected with the vaccine. The documents also reveal Moderna elected not to conduct a number of standard pharmacological studies on the laboratory test animals.

Judicial Watch obtained the records through a September 2021 Freedom of Information Act (FOIA) lawsuit filed after the FDA, the Centers for Disease Control and Prevention (CDC) and the National Institute for Allergy and Infectious Disease (NIAID) failed to respond to a June 2021, FOIA request biodistribution studies and related data for the Pfizer, Moderna and Johnson& Johnson COVID vaccines (Judicial Watch v. U.S. Department of Health and Human Services (No. 1:21-cv-02418)).

The records include a “Nonclinical Overview” prepared by Moderna and submitted to the FDA for approval of its vaccine revealing that a number of rats were born with skeletal deformations, known as “wavy ribs” and “rib nodules,” to mothers injected with the mRNA vaccine. On December 4, 2019 ASPR Robert Kadlec said he'd have an announcements pertaining to newer vaccine manufacturing methods coming up but he couldn't go into particulars



This slide from a presentation to Afrigen by Dr Barney Graham of NIAID says they were discussing a manufacturing demonstration with Moderna in Sept - Nov 2019 - months before the Wuhan flu arrived on Dec 31, 2019.

Graham signed an amendment to the Moderna Research Collaboration on Jan 13, 2020. The RCA on MERS and Nipah was itself signed in May and June 2019. It's obvious this was an orchestrated plan months before any unusual pneumonias

Moderna's Patent clearly shows that DNA fragments in drug products may cause cancer.

Title of Invention
"REMOVAL OF DNA FRAGMENTS IN MRNA PRODUCTION PROCESS"

It's crystal clear from this title that removal of residual DNA fragments in mRNA products is essential.

Interestingly, one of the inventors of this patent is Stéphane Bancel, CEO of Moderna.

CEO of Moderna clearly states, in this patent:

"The DNA template used in the mRNA manufacturing process must be removed to ensure the efficacy of therapeutics and safety, because residual DNA in drug products may induce activation of the innate response and has the potential to be oncogenic in patient populations."

Even the CEO of Modela admits that DNA fragments in mRNA products may cause cancer.
